Centromere protein A (CENP-A) is a heritable epigenetic mark that determines centromere identity and is essential for the accurate segregation of chromosomes during cell division. Here I will discuss the role of DNA methylation in maintaining centromere and neocentromere position, size, and function, and how CENP-A overexpression drives genome instability and oncogenic epigenetic reprogramming during tumor progression.=0D=0A=
